动物源提取法和基因工程重组法各有优缺点。动物源提取法工艺和技术较为成熟,成本相对较低,能够最大程度保留天然胶原蛋白的三螺旋结构,产物生物活性较高。但动物源提取法存在一定隐患:1)异体胶原蛋白可能导 致免疫排斥或过敏反应;2)动物组织提取法产物通常是混合物,未能完全清除的核酸、杂蛋白、多糖等物质亦属于免疫原范畴;3)存在动物源疾病感染 风险,如疯牛病、禽流感等。基因工程法较好克服了免疫原及病毒风险:1)由于不 使用动物源材料,无动物源病毒感染风险;2)可以使用人胶原蛋白编码序列以提高产物与人体的生物相容性,通过序列设计降低免疫原性过敏风险。但重组胶原蛋白存在不容易形成三螺旋结构的缺点,主要系发酵工程中常用的大 肠杆菌和酵母菌因缺乏脯氨酸羟化酶无法生成羟脯氨酸,而羟脯氨酸的羟基是胶原蛋白形成稳定三螺旋结构的关键。
