主流的胶原蛋白提取和制备方法包括动物源提取和基因工程法两大类,动物源提取相对成熟,但存在一定限制,以基因工程技术生产重组胶原蛋白为目前学界及业界研究的重点。动物源胶原蛋白的提取主要通过酸法和酶法进行产业化制备,成本相对更低,但同时具备一定局限性:(1)存在动物源疾病感染风险;(2)异体胶原蛋白可能导致免疫排斥或过敏反应;(3)产能限制;(4)提取物往往是不同分子量胶原蛋白的混合物,纯度难以把控,延伸应用受限;(5)动物源性胶原蛋白变性温度为 40 度,储存运输难度大。以基因工程技术生产重组胶原蛋白为目前学界及业界研究的重点。重组胶原蛋白是通过基因工程技术,将人体胶原蛋白基因进行特定序列设计、酶切和拼接、连接载体后转入工程细胞,通过发酵表达生产胶原蛋白。该方法得到的产物具有安全性好、加工性强、质量稳定等优点,但技术难点在于如何实现菌种、酵种的高密度表达、保证生物活性以及对重组蛋白的分离纯化。现阶段仅有少数企业能够通过基因工程技术量产,代表企业有巨子生物和锦波生物。
